CJC-1295: Compound Reference for Researchers

A neutral reference sheet for CJC-1295, a modified analogue of the first 29 residues of human growth hormone-releasing hormone, in its two common forms (with and without “DAC”): structure, literature context and analytical notes.

For laboratory research use only. This page is educational. It does not describe uses, doses or effects in humans or animals, and nothing here is a recommendation. Our products are not for human or veterinary use.

NamesCJC-1295 (with DAC); CJC-1295 without DAC, also called Modified GRF(1–29) or tetrasubstituted GRF(1–29)
ClassAnalogue of human growth hormone-releasing hormone (GHRH) residues 1–29; 29-residue peptide amide
Core sequence (without DAC)YADAIFTQSYRKVLAQLSARKLLQDILSR-NH2, with D-alanine at position 2. Compared with native GHRH(1–29), four residues are substituted: D-Ala2, Gln8, Ala15 and Leu27.
The “DAC” formAdds a C-terminal lysine carrying a maleimidopropionyl group (“drug affinity complex”), a reactive group designed to bind albumin (reference 5)
Formula and mass (without DAC)C152H252N44O42; ≈ 3367.9 g/mol (average); monoisotopic mass ≈ 3365.89 Da
Form suppliedLyophilized powder
Regulatory statusSupplied for laboratory research only; not an approved drug product.

Sequence and substitutions are as described in the literature and reference databases. The formula calculated from the core sequence matches the formula listed for the no-DAC peptide. Identifiers and masses listed by suppliers for the DAC form vary, so rely on the laboratory’s own mass result for the lot you hold.

What it is

Native GHRH is a 44-residue hormone; its first 29 residues carry most of its activity. CJC-1295 is built on that 29-residue region with four amino-acid substitutions that make it more stable, including a D-alanine at position 2. The version without DAC is the 29-residue peptide itself. The version with DAC has an extra lysine at the C-terminus modified with a maleimidopropionyl group, which is designed to form a covalent bond with the blood protein albumin (reference 5). The two forms are different molecules with different properties and should not be treated as interchangeable.

What the published literature looks like

Published work includes early human studies of hormone responses to a long-acting GHRH analogue (references 1 and 2), animal studies (reference 3), a study of serum protein changes (reference 4) and the original description of the albumin-binding design (reference 5). Because “CJC-1295” is used for both forms, check which form a study used.

These are cited to show the breadth of the literature, not to endorse any conclusion in it.

How to read the evidence

  • Know which form was studied. Findings for the DAC form do not automatically apply to the no-DAC peptide, and the reverse.
  • Research material is not a clinical product. Studies describe specific preparations under controlled conditions. They are not findings about research-use supplies.
  • Quality matters. A 29-residue synthesis has more room for related impurities than a short peptide, so purity and identity data deserve a careful look.

Analytical notes

These points follow from the sequence (see our guides to reading a COA and to HPLC and mass spectrometry).

  • Detection wavelength. The peptide contains two tyrosines and a phenylalanine, so it absorbs at 280 nm as well as at 214–220 nm. Our certificates for these products report the chromatogram at 280 nm. Purity values measured at different wavelengths are not directly comparable.
  • Mass check (no-DAC form). Expected multiply charged ions, using average masses, include about m/z 843.0 (4+), 674.6 (5+) and 562.3 (6+). The DAC form has a higher mass because of the added lysine and maleimidopropionyl group.
  • Chemistry. There is no methionine or cysteine, so oxidation is less of a concern than for other peptides. Three glutamines can deamidate (about +1 Da). The peptide is basic (three arginines and two lysines).
  • Reactive group. The maleimide in the DAC form is chemically reactive, so that form needs particular care in handling and in interpreting an analytical result.

Handling and storage

General laboratory guidance for lyophilized peptides applies: store cold, dry, sealed and protected from light, let the vial reach room temperature before opening, and avoid repeated freeze–thaw cycles for any solution. Details are in our guide to research peptide storage and stability.
